The nucleosome-remodeling ATPase ISWI is regulated by poly-ADP-ribosylation.

ATP-dependent nucleosome-remodeling enzymes and covalent modifiers of chromatin set the functional state of chromatin. However, how these enzymatic activities are coordinated in the nucleus is largely unknown. We found that the evolutionary conserved nucleosome-remodeling ATPase ISWI and the poly-ADP-ribose polymerase PARP genetically interact. We present evidence showing that ISWI is target of poly-ADP-ribosylation. Poly-ADP-ribosylation counteracts ISWI function in vitro and in vivo. The GRN specifying the dorsal-ventral (D-V) axis of the sea urchin embryo is currently under investigation. An early input for D-V polarity is given by a redox gradient probably generated by an asymmetrical distribution of maternal mitochondria (1). Only on the future ventral side, the oxidizing environment induces the expression of the nodal gene, an essential regulator of D-V polarization (2). By contrast, on the future dorsal side, a reducing environment activates the hypoxia inducible factor (HIF-1α) (3). Comprehensive analysis of interacting proteins and genome-wide location studies of the Sas3-dependent NuA3 histone acetyltransferase complex

Highlights • We characterise Sas3p and Gcn5p active HAT complexes in WT and deleted TAP-strains. • We confirm that Pdp3p interacts with NuA3, histones and chromatin regulators. • Pdp3p MS-analysis reveals its phosphorylation, ubiquitination and methylation. • Sas3p can substitute Gcn5p in acetylation of histone H3K14 but not of H3K9. • Genome-wide profiling of Sas3p supports its involvement in transcriptional elongation.

Geschlechts-Chromatin und Handleistenmuster in einer Landesnervenklinik

Die 804 mannlichen Patienten einer allgemeinen Landesnervenklinik wurden systematisch nach Kerngeschlecht und Handleistenmustern untersucht. Dabei fanden sich 0,62% chromatin-positive, nach der Chromosomenanalyse verschiedene Typen des Klinefelter-Syndroms. Wenn auch eine Reihe von Papillarleistenmustern fur Aberrationen der Geschlechtschromosomen charakteristisch sind, so ist die Auspragung doch zu variabel, um diese Aberrationen untereinander zu differenzieren. Die Auswahl nach Grose und kriminellen Vorgeschichte forderte in diesem Material keinen YY-Fall zu Tage.

Cyclic AMP-induced Chromatin Changes Support the NFATc-mediated Recruitment of GATA-3 to the Interleukin 5 Promoter

Elevated intracellular cyclic AMP levels, which suppress the proliferation of naive T cells and type 1 T helper (Th1) cells are a property of T helper 2 (Th2) cells and regulatory T cells. While cyclic AMP signals interfere with the IL-2 promoter induction, they support the induction of Th2-type genes, in particular of il-5 gene. We show here that cyclic AMP signals support the generation of three inducible DNase I hypersensitive chromatin sites over the il-5 locus, including its promoter region. Multiple roles for ISWI in transcription, chromosome organization and DNA replication.

ISWI functions as the ATPase subunit of multiple chromatin-remodeling complexes. These complexes use the energy of ATP hydrolysis to slide nucleosomes and increase chromatin fluidity, thereby modulating the access of transcription factors and other regulatory proteins to DNA. Here we discuss recent progress toward understanding the biological functions of ISWI, with an emphasis on its roles in transcription, chromosome organization and DNA replication.

The distribution of active RNA polymerase II along the transcribed region is gene-specific and controlled by elongation factors.

In order to study the intragenic profiles of active transcription, we determined the relative levels of active RNA polymerase II present at the 3'- and 5'-ends of 261 yeast genes by run-on. The results obtained indicate that the 3'/5' run-on ratio varies among the genes studied by over 12 log(2) units. This ratio seems to be an intrinsic characteristic of each transcriptional unit and does not significantly correlate with gene length, G + C content or level of expression. The correlation between the 3'/5' RNA polymerase II ratios measured by run-on and those obtained by chromatin immunoprecipitation is poor, although the genes encoding ribosomal proteins present exceptionally low ratios in …

Imitation SWI (ISWI) and other ATP-dependent chromatin-remodeling factors play key roles in transcription and other processes by altering the structure and positioning of nucleosomes. Recent studies have also implicated ISWI in the regulation of higher-order chromatin structure, but its role in this process remains poorly understood. To clarify the role of ISWI in vivo, we examined defects in chromosome structure and gene expression resulting from the loss of Iswi function in Drosophila.
